RPMI 1640 Medium, no glutamine Gibco™

RPMI 1640 Medium was originally developed to culture human leukemic cells in suspension and as a monolayer. Roswell Park Memorial Institute (RPMI) 1640 Medium has since been found suitable for a variety of mammalian cells, including HeLa, Jurkat, MCF-7, PC12, PBMC, astrocytes, and carcinomas. Using RPMI
RPMI 1640 Medium is unique from other media because it contains the reducing agent glutathione and high concentrations of vitamins. RPMI 1640 Medium contains biotin, vitamin B12, and PABA, which are not found in Eagle's Minimal Essential Medium or Dulbecco's Modified Eagle Medium. In addition, the vitamins inositol and choline are present in very high concentrations. RPMI 1640 Medium contains no proteins, lipids, or growth factors. Therefore, RPMI 1640 Medium requires supplementation, commonly with 10% Fetal Bovine Serum (FBS). RPMI 1640 Medium uses a sodium bicarbonate buffer system (2.0 g/L), and therefore requires a 5–10% CO2 environment to maintain physiological pH.

BenchStable™ DMEM Gibco™

Gibco DMEM (Dulbecco's Modified Eagle Medium) is a widely used basal medium for supporting the growth of many different mammalian cells. Cells successfully cultured in DMEM include primary fibroblasts, neurons, glial cells, HUVECs, and smooth muscle cells, as well as cell lines such as HeLa, 293, Cos-7, and PC-12. BenchStable DMEM has been engineered for storage at room temperature while supporting cellular proliferation and maximum cell densities comparable to conventional DMEM when supplemented with ≥10% fetal bovine serum (FBS).

This DMEM is modified as follows:

Using BenchStable DMEM
DMEM is unique from other media as it contains four times the concentration of amino acids and vitamins as the original Eagle's Minimal Essential Medium. DMEM was originally formulated with low glucose (1 g/L) and sodium pyruvate, but is often used with higher glucose levels, with or without sodium pyruvate. DMEM uses a sodium bicarbonate buffer system (3.7 g/L) and so requires a 5–10% CO2 environment to maintain physiological pH. DMEM with GlutaMAX supplement minimizes toxic ammonia build-up and improves cell viability and growth in an easy-to-use format. BenchStable DMEM contains no proteins, lipids, or growth factors and so requires supplementation with ≥10% fetal bovine serum (FBS).

Ready-to-use format
Gibco® AmnioMAX™ - II Complete Medium is a frozen, 1X medium, ready to use upon thawing. No supplementation is required. Thawed medium can be stored at 2–8°C for up to ten days.

Quality and performance testing
Every lot of Gibco® AmnioMAX™ - II Complete Medium is performance tested by a certified US reference cytogenetics laboratory to ensure consistently superior performance. Pooled primary human amniotic fluid samples are cultured for six days in Gibco® AmnioMAX™ - II Complete Medium before measuring the total number of colonies and total number of mitotic colonies. In addition, each lot is tested for sterility, pH and osmolality.

Unique, optimized formulation
Gibco® AmnioMAX™ - II Complete Medium contains Fetal Bovine Serum (FBS), gentamicin, and L-glutamine to maximize cell attachment and growth. This optimized medium also has an enhanced buffering system that provides greater pH stability during culture manipulations.

Neurobasal Medium, minus phenol red is a basal medium designed for long-term maintenance and maturation of pure pre-natal and embryonic neurons without the need for an astrocyte feeder layer when used with Gibco B-27 supplements. Use this modified formulation of Neurobasal Medium when the presence of phenol red will interfere with your results.

Neurobasal Medium is suitable for use in most neuronal cell applications. For other applications, we provide alternative formulations of Neurobasal media and B-27 supplements.

B-27™ Supplement (50X), minus antioxidants Gibco™

A variation of the most cited neuronal cell culture supplement, B-27 Supplement Minus Antioxidants (AO) is a customized version of the original B-27 Supplement without five antioxidants (vitamin E, vitamin E acetate, superoxide dismutase, catalase, and glutathione). B-27 Supplement Minus AO is a serum-free supplement designed for studies of the role of free-radicals in aging, toxicity, apoptosis, and chronic neurologic diseases. B-27 Supplement Minus AO is provided as a 50X liquid and is intended for use with Neurobasal Medium or Neurobasal-A Medium for neuronal cell culture without the need for an astrocyte feeder layer.

B-27 Supplement is suitable for use in most neuronal cell applications. For other applications, we provide alternative formulations of B-27 Supplement and Neurobasal media.

Keratinocyte SFM (1X), without calcium chloride Gibco™

Keratinocyte SFM (serum-free medium) is optimized for the growth and maintenance of human keratinocytes and other types of epithelial cells without the need for a feeder layer of cells. The complete medium may be used to cultivate human epidermal keratinocytes for studies involving dermal substitutes, in vitro toxicology, and gene therapy. Keratinocyte SFM without calcium is available to allow researchers to adjust the calcium concentration to meet the demands of their specific application.

Keratinocyte SFM is a complete serum-free medium when supplemented with human recombinant epidermal growth factor (rEGF) and bovine pituitary extract (BPE) at the time of use. Both supplements, rEGF and BPE, are supplied in separate packaging with the basal medium.

Viral Production Cells Gibco™

Gibco Viral Production Cells are a derivative of the HEK 293F cell line and are a core component of the LV-MAX Lentiviral Production System. Viral Production Cells have been adapted to a special chemically-defined, serum-free and protein-free LV-MAX Production Medium. They are maintained in suspension culture and will grow to high density in LV-MAX Production Medium. Viral Production Cells are highly transfectable by LV-MAX transfection reagent and generate superior lentiviral production compared to polyethyleneimine (PEI)-based lentiviral production methods.

• Suspension culture cells facilitate high-throughput screening or scale-up

Frozen cells are supplied in a vial containing 1 mL of cells at 1 x 107 viable cells/mL in LV-MAX Production Medium and 10% DMSO. The cells should be thawed directly into LV-MAX Production Medium.

The cells are typically grown in flasks on a shaker platform in a humidified, CO2 cell culture incubator. However, cultures are scalable in volume from 96-deep well blocks to 2-L flasks and bioreactors. Lentiviral vector harvest is typically performed at two days post-transfection.

Using DMEM/F-12

EfficientFeed™ B+ AGT™ Supplement Gibco™

Gibco® EfficientFeed™ B+ (Plus) AGT™ Supplement is a dry-format, single-part supplement for fed batch addition to the culture of multiple cell lines to assist with process development and maximization of bioreactor utilization. Compared to original CHO CD EfficientFeed™ B AGT™ nutrient supplement, EfficientFeed™ B+ supplement has components that are uniquely combined to allow simpler single-part pH-neutral reconstitution, as well as the ability to super-concentrate the supplement from 52.7 g/L (1X) to 158.1g/L (3X) for reduction of product dilution or increased titers with additional feeding, when appropriate. The product is an animal origin-free, chemically defined formulation that contains no proteins, hydrolysates, or components of incompletely defined composition.

Consider EfficientFeed™ B+ AGT™ Supplement when:

Increasing Titers
As a complete feed, EfficientFeed™ + supplements have been shown to provide multiple-fold increases in titers at 1X concentration versus batch culture, and are an excellent starting point for process development. Flexibility to further increase titers are enhanced with the ability to super-concentrate the product by reconstituting with less water to deliver up to 158 g/L of fed nutrients (see figure).

Maximizing Bioreactor Utilization
Have you ever wished you could have more working volume at the start of a bioreactor run to get more protein produced, but needed too much feeding volume space to accomplish your optimal goal? Have you ever thought that room for a little more feeding might get you to a new level of titer from a process and maybe even reduce the number of bioreactor runs that might be needed per year? In either of these cases, it is important to have the flexibility to super-concentrate your feeding supplements so you get less product dilution, more space for an additional feeding, and/or more working volume to start your run (see figure). In fact, you may gain about 20% more bioreactor utilization by exploring the unique ability to super-concentrate this EfficientFeed B+ product.

Simplified Handling to Reduce Risk in Scale-Up
The ability to scale up in an AGT™ dry format provides ease of handling and storage in manufacturing and other bioprocess settings. Fewer parts and steps in reconstitution mean fewer opportunities for operator error. Further, ability to store this feed, even when super-concentrated, for the length of a bioreactor run at room temperature, in the dark, may allow you fewer chances of contamination during closed system connections by reducing the number of connections to the bioreactor to one.

DMEM, high glucose, pyruvate Gibco™

StemPro™-34 SFM (1X) Gibco™

StemPro®-34 SFM is a serum-free medium specifically formulated to support the development of human hematopoietic cells in culture (1). StemPro®-34 SFM can be used to study hematopoietic cells from bone marrow, peripheral blood, or neonatal cord blood. StemPro®-34 SFM is performance-tested on normal human bone marrow. It is manufactured with components of human, recombinant or synthetic origin.

StemPro®-34 SFM consists of a basal liquid medium and frozen supplement (StemPro®-Nutrient Supplement) which is added at the time of use.
